ثبت نشده
چکیده
Metabolic conversion of oestrogen phenolic groups may create conditions of oxidative stress accompanied by damage of cellular macromolecules including DNA. The aim of this investigation was to evaluate the cell cycle kinetics and possible cytogenetic changes in cultured human peripheral blood lymphocytes exposed to seven experimental concentrations of 17 -oestradiol (range 10 M to 10 M). Cell cycle kinetics was analysed on metaphase spreads prepared for a standard analysis of sister-chromatid exchanges (SCEs) stained by fluorescent-plus-Giemsa (FPG) technique. Cytogenetic changes were monitored by analysis of chromosome damage (gaps and breaks), structural and numerical aberrations. On the basis of the obtained results it can be concluded that oestradiol has no significant influence on cell cycle kinetics and mitotic index of cultured human lymphocytes. However, at estradiol concentration of 7×10 M, and at higher concentrations used in this experiment, there was a significant increase of gaps, breaks and aneuploidies. On the other hand, oestradiol treatment has not changed the frequency of polyploid cells. Therefore, it can be concluded that high concentrations of oestradiol pose some genetic risk detectable at cytogenetic level.

Application of nuclear magnetic resonance imaging (NMRI) as a non-invasive and accurate imaging procedure has been widely used in recent years. Meanwhile, the biological effects of magnetic fields of several tesla (T) and high energy radiofrequency (RF) is not fully known yet. Because of controversy over this issue, the present research has been carried out in order to verify the effects of...

Background: A radioprotective effect of amifostine as well as its ability to modulate the level of spontaneous and gamma-irradiation-induced genetic changes on human peripheral blood lymphocytes has been investigated . Amifostine, known as a potent radical scavenger, has been introduced as the most effective radioprotector, yet it is not completely approved for the clinical use. However, furt...
متن کاملEffect of Myeloma-Derived Microparticles on in Vitro Proliferation and Viability of Alloimmune Human Peripheral Blood Mononuclear Cells
Background: Microparticles (MPs) are small vesicles released from the cell membrane. Accordingly, these contain active molecules to mediate biological processes, including cell proliferation and cell cycle progression. The fusion of myeloma cell lines with immunized B lymphocytes is a critical step of hybridoma technology. MPs modulate lymphoproliferation, thereby facilitating B cell expansion ...

Objective(s): Chlorpyrifos (CP) is a broad-spectrum organophosphorus pesticide used extensively in agricultural and domestic pest control, accounting for 50% of the global insecticidal use. In the present study, protective effects of two selenium-enriched strong antioxidative medicines IMOD and Angipars were examined in human lymphocytes treated with CP in vitro. Materials and Methods: Isolate...
